The ubiquitin-proteasome system (UPS) plays key roles in our cellular system by participating in regulating protein quality control and general protein homeostasis. As an irreplaceable component, the E3 ubiquitin ligase also functions in proteolysis targeting chimeras (PROTACs) with the help of E3 ligase ligands. However, only a limited number of E3 ubiquitin ligases have been successfully used in this high-ending technology: the Von Hippel-Lindau disease tumor suppressor protein (VHL), the Mouse Double Minute 2 homologue (MDM2), the Cellular Inhibitor of Apoptosis (cIAP), and cereblon. Since more than 600 E3 ubiquitin ligases in human proteome have been identified, it’s great potential to try more new ligases in the PROTAC molecule development. Creative Biolabs has produced several E3 ubiquitin ligases and customers could conduct in vitro function tests of their preliminary PROTAC molecules. We are also working to develop more E3 ubiquitin ligases by incorporating our cell-free protein expression system.
